چکیده

Age-related changes in al-, a?-, and 0-catecholamine receptors on membranes of rat epididymal fat cells were investigated. Both young (6 weeks old, weight about 190 g) and aged (20 weeks old, weight about 490 g) Sprague-Dawley male rats were used. For the a]-adrenoceptor binding experiment, we developed a novel analytical method using the hydrophilic a]receptor selective antagonist, [3H]bunazosin. The binding of [3H]bunazosin to its binding sites was rapid, reversible, saturable, and stereospecific. Scatchard binding analysis showed a single class of binding site. The sites were characterized as a]adrenoceptors by inhibition experiments using various agonists and antagonists. The number of maximum binding sites (Bmm) of al-receptor binding was 37.0 f 6.5 (young) versus 24.0 f 3.2 (aged) fmol/mg protein (P < 0.01). [3H]Rauwolscine and [3H]CGP-12177 were used for a2and j3-receptor binding, respectively. In a2-receptor detection using [3H]rauwolscine as a ligand, B,, increased markedly from 19.8 + 4.9 to 86.2 + 19.5 fmol/mg protein (P < 0.01). In contrast, B,,, for @-receptor decreased from 69.7 + 9.7 to 45.4 + 13.9 fmol/mg protein with increasing rat age (P < 0.05). Kd showed no change in each of the binding experiments between young and aged rats. The cell volume increased from 0.07 & 0.02 to 0.15 * 0.06 nl. It is implied that anti-lipolytic activity strengthened on the whole mainly with the marked increase of a2-receptor number and decrease of j3-receptor number. Furthermore, [3H]bunazosin was demonstrated to be a new suitable radioligand for the detection of the al-adrenoceptor of rat white fat cell membranes.-Kobatake, T., Y. Watanabe, Y. Matsuzawa, K. On the other hand, catecholamines also perform an anti-lipolytic role by stimulating az-adrenoceptors and inhibiting adenylate cyclase (1). The existence of at-adrenoceptors, which could not be shown owing to the impossibility of direct binding of [3H]prazosin (2) on rat white fat cell membranes, has been suggested indirectly by the demonstration of responses to al-adrenoceptor agonists such as phosphatidylinositol turnover or Caz + mobilization (3), although their physiological roles in white fat cells have not been

Immunomodulating Effect of Cyclophosphamide on the Mice Infected with Candida albicans

Background & Objectives: Cyclophosphamide is an alkylating agent that stops the replication of DNA, which is used to treat various types of cancer and some autoimmune disorders. This study was aimed at then evaluating the immunomodulating effect of cyclophosphamide (Cy) on the immune system of vaccinated and non-vaccinated mice.Materials & Methods: The  study was performed on three groups of mi...

Global gene expression profiling in early-stage polycystic kidney disease in the Han:SPRD Cy rat identifies a role for RXR signaling.

Han:SPRD Cy is a spontaneous rat model of polycystic kidney disease (PKD) caused by a missense mutation in Pkdr1. Cystogenesis in this model is not clearly understood. In the current study, we performed global gene expression profiling in early-stage PKD cyst development in Cy/Cy kidneys and normal (+/+) kidneys at 3 and 7 days of postnatal age. Expression profiles were determined by microarray...

Altered ATP-sensitive P2 receptor subtype expression in the Han:SPRD cy/+ rat, a model of autosomal dominant polycystic kidney disease.

The effects of extracellular ATP on fluid secretion and reabsorption by renal epithelial cells, as well as its known effects on cell proliferation and death, are potentially important contributory factors in the development and growth of renal cysts. In this study, we have investigated the protein and mRNA expression of several P2Y receptor subtypes (P2Y(1,2,4,6)), as well as the P2X(5) and P2X...

Improvement of Large-scale PRP production by Haemophilus influenzae typeb, using modified CY medium

Background and Objective: Haemophilus influenzae type b (Hib) is a gram negative bacterium and one of the most common causative agents of acute meningitis in infants and less than 5 years old children worldwide. The production of Hib capsular polysaccharide polyribosyl ribitolphosphate (PRP) is important for the production of conjugate vaccines against Hib infections. The aim of this study is t...

Protective effect of Cy-3G on PC12 cells against beta-amyloid-induced apoptosis and the possible mechanism

Beta-amyloid protein (Abeta), a major protein component of brain senile plaques in Alzheimer’s disease, is known to be directly responsible for the production of reactive oxygen species (ROS) and induction of apoptosis. In this study, the protective effect of cyanidin 3-O-glucopyranoside (Cy-3G), an anthocyanin derivate purified from fruits, on Abeta-induced rat pheochromocytoma (PC12) cultures...
